Preparation of Cell Wall Degrading Enzymes from Marine Herbivores for Algal Protoplast Isolation 1. Cell Wall Degradation and Isolation of Protoplasts from a Marine Red Alga Porphyra okamurae Ueda by Enzyme Extracts of Abalone (Nordotis discus (Reeve))

ABSTRACT
In order to develop algal cell wall degrading enzymes, for the protoplast colation, from marine herbivores native to Korean coast, the acetone powder extract of abalone (Nordotis discus (Reeve)) was prepared. Protoplasts were isolated from a marine red alga Porphyra okamurae Ueda using prepared acetone powder extracts of abalone and commercial cellulase mixture. The optimal condition for the release of protoplasts was found to be a combination of 1% prepared abalone acetone powder and 3% cellulase in 0.5M mannitol at pH 6.0. The acetone powder extract of abalone produced similar or better protoplast yield compared to the commercial abalone acetone powder.
